Objectives:Toinvestigatethedistributionandfunctionofolfactoryensheathingcells(OECs)followinglumbarpuncture(LP)transplantationinmousespinalcordinjury(SCI).
Methods:OECsweretransplantedbyLPatlevelL3¨C5,1weekaftertransectedSCIatT8vertebra.Micewerekilledat3,21,and56daysafterLPtransplantation,andtherelativedistributionofcellsatT8vertebrawasquantitated.Theinjuredspinewasalsotestedbyimmunohistochemistrytoassessneuronregenerationandscarbridgingat8weeksposttransplantation.MotorfunctionsofmicewereevaluatedduringtheobservationperiodusingtheBassoMouseScale.
Results:OECswereexaminedandconfirmedbystudyingcellmorphologyunderphasecontrastandimmunostainingofNGFRp75.LP-transplantedOECscouldbedetectedjust3daysaftertransplantation(p75+area:0.16mm2)andaccumulatedto0.31and0.30mm2at21and56dayspostengraftment,respectively.Thenumberofendogenousneurons,?to+¦Ìm,farfromtheepicenter,inOEC-transplantedmicewasmorethanthatinSCImicewithoutengraftment.SCIlesionofmiceinthecontrolgroup(.3¡À3.97¦Ìm)wasmuchlongerthanthatinOEC-graftedgroup(.7¡À3.60¦Ìm).GraftsofOECsinducedsignificantfunctionalimprovementinmicethatunderwentT8vertebraltransection,justfrom3daysaftercellinjection.
Conclusion:LPisaminimallyinvasivemethodforOECtransplantationtotreatSCI.ThisisthefirststudytovisualizethedistributionandfunctionsofLP-transplantedOECsintheintactandinjuredspinalcord.
